Crystalline activated protein B (chymotrypsin B).

چکیده
The protein isolated from beef pancreas (1) was recently identified as the zymogen of a proteolytic enzyme and provisionally called protein B (2). In this paper a method of crystallization of the activated enzyme is described. Further properties of this enzyme have been studied, suggesting that the activated protein B belongs to the group of chymotrypsins. The strongest evidence for this conclusion came from the work of Dr. Fruton (3) in which the specificity of the activated protein B toward several synthetic peptides has been investigated. The activity was found to be similar to that of the group of chymotrypsins. In view of our own, and particularly Dr. Fruton’s results, we have proposed the name chymotrypsin B for the active enzyme and chymotrypsinogen B for the unactivated form.

Specificity of chymotrypsin B.
Brown, Shupe, and Laskowski (1) have described the isolation, from beef pancreas, of a crystalline proteolytic enzyme which they have named “activated protein B” or “chymotrypsin B.” Through the kindness of Dr. Laskowski who provided us with a 4 times recrystallized sample of this enzyme, it was possible to examine its action on a series of synthetic peptides and peptide derivatives. The data p...
متن کاملProteases Detection of invitro Culture of Midgut Cells from Hyalomma anatolicum anatolicum (Acari: Ixodidae)
Proteases play a key role in protein digestion in ticks and other haematophagous insects. Our understanding of blood meal digestion in digestive system of ticks can be very useful for better understanding of basic rules for control of ticks. Cells of the midgut endocytose blood components. Blood proteins uptake by midgut cells, suggesting the presence of proteases in the midgut cells. In this...

Extracts of the body walls of Ascaris lumbricoides were reported by Green (2) to contain one substance capable of inhibiting chymotrypsin cy and another substance capable of inhibiting trypsin. Peanasky and Laskowski (3) separated these inhibitors by means of continuous paper curtain electrophoresis and crystallized the chymotrypsin inhibitor. The crystalline preparation, however, was heterogen...
متن کاملAnti-inflammatory Effects of Oxymatrine Through Inhibition of Nuclear Factor–kappa B and Mitogen-activated Protein Kinase Activation in Lipopolysaccharide-induced BV2 Microglia Cells
Oxymatrine, a potent monosomic alkaloid extracted from Chinese herb Sophora japonica (Sophora flavescens Ait.). possesses anti-inflammatory activittyes. This study was designed to investigate the effects of oxymatrine on nuclear factor–kappa B (NF-κB) and mitogen-activated protein kinase (MAPK)-dependent inflammatory responses in lipopolysaccharide (LPS)-activated microglia. In this paper, BV2...
